چکیده

A major challenge in the field of RNA chemistry is identification selective and quantitative conversion reactions on that can be used for tagging any other tool development. Here, we introduce metal-free diazotransfer native containing an aliphatic primary amino group using diazotizing reagent fluorosulfuryl azide (FSO2N3). The reaction provides corresponding azide-modified nearly quantitatively yields without affecting nucleobase groups. obtained azido-RNA then further processed utilizing well-established bioortho-gonal reactions, such as azide-alkyne cycloadditions (Click) or Staudinger ligations. We exemplify robustness this approach synthesis peptidyl-tRNA mimics pull-down 3-(3-amino-3-carboxypropyl)uridine (acp3U)- lysidine (k2C)-containing tRNAs Escherichia coli tRNA pool isolated from cellular extracts. Our therefore adds a new dimension to targeted chemical manipulation diverse species.

Relation Between RNA Sequences, Structures, and Shapes via Variation Networks

Background: RNA plays key role in many aspects of biological processes and its tertiary structure is critical for its biological function. RNA secondary structure represents various significant portions of RNA tertiary structure. Since the biological function of RNA is concluded indirectly from its primary structure, it would be important to analyze the relations between the RNA sequences and t...

Enhancement of RNA Interference Effect in P19 EC Cells by an RNA-dependent RNA Polymerase

Background: RNA interference (RNAi) is a phenomenon uses double-stranded RNA (dsRNA) to specifically inhibit gene expression. The non-specific silencing caused by interferon response to dsRNA in mammalian cells limits the potential of utilizing RNAi to study gene function. Duplexes of 21-nucleotide short interfering dsRNA (siRNA) inhibit gene expression by RNAi. In some organisms, siRNA can als...
